Splenocyte subpopulations were purified by combined positive and negative selection 2 weeks after completion of immunization or 4 weeks after infection; the frequencies of hsp65-reactive cells were then assessed by limiting dilution analysis with J774-hsp65 as antigen-presenting cells.
