Adoptive transfer of Toxoplasma antigen-responsive CD8⁺ T cells. CD8⁺ T cells were isolated by magnetic bead separation from the splenocytes of Neospora-vaccinated mice (Neo). The purified CD8⁺ T cells were 95% pure as determined by FACS and were cultured in vitro in the presence of the Toxoplasma antigen. Five days postculture, the proliferating CD8⁺ T cells were recovered (>98% purity as determined by FACS) and adoptively transferred (10⁷ cells) via tail vein injection into naive mice. Mice were challenged with 10⁴ PLK strain tachyzoites.
