Model of AR and FOXA1 mediated repression of NR3C1 expression. The expression of NR3C1 is repressed by AR through EZH2, and by FOXA1 through TLE3. Inhibition of AR by ENZ and/or depletion of FOXA1 by siRNA (siFOXA1) leads to de-repression of NR3C1. De-repression of NR3C1 leads to increased GR protein levels, and augmentation of GR transcriptional activity. GR occupies AR binding sites at pre-accessible chromatin sites enriched with coregulator proteins.
