Glycosylation of RhCMV US6 family proteins. HeLa cells were transiently transfected with HA-tagged versions of the US6-related genes of RhCMV and labeled with [35S]cysteine-[35S]methionine for 30 min, followed by a 60-min chase where indicated. US6 family proteins were immunoprecipitated from 1% NP-40 lysates with monoclonal anti-HA antibody (Sigma). The immunoprecipitates were either left untreated (−) or treated (+) with EndoH prior to sodium dodecyl sulfate-polyacrylamide gel electrophoresis and autoradiography.
