Isogenic stt3-7 single- and double-mutant cells were plated in serial dilutions on rich medium and grown at 23 and 30°C for 3 days. (B) Overexpression or deletion of the UBC6 gene stabilized the mutated Stt3-7 protein. Cells were shifted to 37°C and protein was extracted. Steady-state protein levels and CPY glycosylation were determined by western analysis using the indicated antisera. Hexokinase was used as the control for equal protein loading.
