Tetracycline-regulated expression of Flag-tagged CKIP-1. Flag-tagged CKIP-1 is expressed in the cell line DC1.4 under the tight control of tetracycline (tet) as shown by immunoblotting (A) and immunofluorescence (B). (A) DC1.4 cells were grown in the presence or absence of tetracycline for 24 h to induce the expression of Flag-CKIP-1. Extracts were prepared, and 35 μg of total-cell lysate was separated by SDS-PAGE, transferred to a polyvinylidene difluoride membrane, and immunoblotted with anti-Flag M2 antibodies. (B) DC1.4 cells grown in the presence or absence of tetracycline were fixed, permeabilized, and stained with anti-Flag M2 antibody and TRITC-goat anti-rabbit antibody.
