The method for assessing pseudopod frequency can be found in Wessels et al. (47). Frequencies are provided as means ± standard deviations. Individual cells (total numbers of cells examined are given in parentheses) were analyzed for 10-min periods. P values were computed by a Student's t test; a P value of >5 × 10−2 was considered nonsignificant. P values for the comparison of JH10 cells with acaA and acaA (r) cells were nonsignificant in buffer; in spatial gradient, the P values were 6 × 10−8 and 2 × 10−6, respectively, and in front of the temporal wave, they were 4 × 10−6 and 2 × 10−3, respectively. In buffer versus spatial gradient, the P values were 4 × 10−5 for JH10 cells and nonsignificant for acaA and acaA (r) cells. In buffer versus the front of the temporal wave, the P values were 1 × 10−4 for JH10 cells, 1 × 10−2 for acaA cells, and 6 × 10−2 for acaA (r) cells.
