Pheromone-induced growth arrest of S. cerevisiae transformants expressing the S. macrospora pre2 pheromone receptor gene. (A) Quantitative halo test of MATa lawns (7416-12-3; Δste2 sst2-1) either expressing the S. macrospora pre2 pheromone receptor gene (+PRE2) or carrying the mock vector pPGK (+pPGK). Sterile filter disks contained 20 μl of cell-free culture supernatant of MATα cells expressing (+PHE) or not expressing (−PHE) the S. macrospora ppg1 gene. (B) Response of MATa lawn (strain 7416-12-3) expressing the S. macrospora PRE2 receptor to a single dose of synthetic pheromone PHE1 (1.5 nmol/disk), α-factor (1.5 nmol/disk), and 2% dimethyl sulfoxide (DMSO; 10 μl/disk).
