In vitro labeling of minus-strand strong-stop DNA. Wild-type (WT) and B0_GB1_G mutant VLPs were incubated with all four deoxyribonucleotide triphosphates, [α-³²P]dATP, and [α-³²P]dTTP under conditions that allowed for RT. Reaction products were deproteinized and run on a DNA sequencing gel. A sequencing reaction (lane M) was used to generate size markers (sizes are indicated in nucleotides [nt]). A 171-base species is observed with wild-type VLPs, whereas this molecule is not observed with B0_GB1_G mutant VLPs.
