Total RNA was extracted from cells cultured in the presence or absence (∼12 h) of IL-3 and subjected to Northern blot analysis of cdc2 and cdk2 expression as described in Materials and Methods. 28S rRNA stained with methylene blue is shown. Membranes were reprobed following dehybridization. (B) Transcriptional analysis of the cdc2 gene in BMC and BAF-B03 cells under different conditions. cdc2 gene plasmids bound to nitrocellulose were hybridized with 32P-labelled run-on transcripts from nuclei isolated from BMC and BAF-B03 cells IL-3 stimulated or IL-3 deprived for 12 h.
