AGO2 subcellular localization depends on environmental conditions, availability of miRNAs, and expression of miRISC cofactor, TNRC6. Summary scheme showing that multiple variables can affect nuclear localization of AGO, including growth to high density in 2D cell culture, knockout of DROSHA, knockout of DROSHA combined with introduction of exogenous small RNA, knockout of TNRC6, attachment of an NLS sequence to AGO, HCT116 3D cultured to form tumor spheroids, and in human colon tumor tissue.
