AiV-A1 isolate kvgh99012632/2010 cell entry is dynamin- and cholesterol-mediated and strongly dependent on clathrin and endosomal acidification. (A–E) A549 cells were preincubated with the indicated compounds for 30 min at 4 °C and AiV-A1 kvgh99012632/2010 was added (MOI=1) for additional 30 min at 4 °C. The cells were then transferred to 37 °C for 8 h, lysed and the vRNA was measured by RT-qPCR. (F) A549 were pre-treated for 30 min at 4 °C with the indicated compounds, followed by another incubation for 1 h at 37 °C together with the virus and its subsequent removal by washing with PBS containing NH₄Cl. The cells were then incubated for another 8 h at 37 °C, lysed and the vRNA was measured by RT-qPCR.
