Inhibition of TLR3 signaling by the W protein is dependent on its nuclear localization. Activation of the ISG54 promoter by overexpression of TRIF was performed as described in Fig. 4B. The V-SV40 NLS and WBR3-SV40 NLS constructs contain the NLS from the SV40 T antigen at the extreme C terminus of the V and WBR3 ORFs, respectively. The subcellular localization of the proteins encoded by the V and W expression constructs is indicated above the corresponding bar for fold activation.
