CEC were transfected with mutant DNA to reconstitute virus. Infected cells were then used to inoculate fresh CEC or SOgE cells. The respective (mutant) virus is indicated. As primary antibodies for indirect immunofluorescence analysis, a polyclonal MDV-specific chicken serum (α-MDV serum) or a gC-specific rabbit antiserum (α-gC serum) were used. Infected cells and expression of gC were detected with Alexa Fluor 488 goat anti-chicken IgG and Alexa Fluor 568 goat anti-rabbit IgG as secondary antibodies. Plaques were photographed with a fluorescence microscope (Zeiss Axiovert 25) and a digital camera (Zeiss Axiocam).
