Model of the role of interactions of fusion protein AML1/ETO with the N-CoR–SMRT–Sin3–HDAC complex. (a) DNA-bound AML1 interacts with p300 and potentially with pCAF and nuclear receptor coactivators (CoA). This association results in increased histone acetylation, chromatin remodeling, and other effects on the transcriptional machinery resulting in transcriptional activation. (b) AML1/ETO recruits the N-CoR–Sin3–HDAC complex, decreasing histone acetylation and producing repressive chromatin organization and other effects on the transcriptional machinery resulting in transcriptional repression of some genes and, potentially, activation of a subset of other genes.
