PCR analysis to evaluate the proviral copy number after retroviral transduction of PBMC from an ADA⁻ SCID patient. PCR amplification with the sense and antisense primers indicated in the left panel yields two bands in cells containing proviral integrants: a 491-bp band from the endogenous ADA gene and a 240-bp band from the proviral integrant. The ratio values were determined by dividing the phosphoimager signal of the vector DNA band by that of the genomic DNA band.
