(A) The Rep78 and VP2/3 ORFs were cloned into the pFastBacDual vector under the control of the polyhedrin (PPH) or p10 (Pp10) promoters, respectively. (B) The Rep52 and VP1 ORFs were cloned into the pFastBacDual vector under the control of the polyhedrin (PPH) or p10 (Pp10) promoters, respectively. The ITR-containing GOI was cloned adjacent to the Rep52 ORF. (C) The AAV6 VP1 sequence was modified to contain the AAV2 phospholipase A2 (PLA2) domain and a short fragment of VP1 from AAV9 at the indicated nucleotide coordinates.
