Analysis of the binding site size of I3 on ssDNA. Reaction mixtures contained either 10.5, 1.5, or 0 pmol of purified recombinant I3 and 0.2 pmol of a terminally radiolabeled oligonucleotide of 18, 24, 34, 53, or 73 nt in length. After 15 min of incubation at 30°C, samples were fractionated on a nondenaturing, 5 to 20% polyacrylamide gradient gel at 4°C and visualized by autoradiography.
