IL-12 p40 mRNA expression by sorted monocytes/macrophages. Monocytes/macrophages were separated by use of magnetic beads. Mouse anti-bovine CD14 antibody was used, followed by goat anti-mouse IgG-coated magnetic beads. Sorted monocytes/macrophages were confirmed by esterase staining. β-actin was used to confirm the RT-PCR results and that a similar product was evident in different animals and isolated cell populations.
