Mutant TNS1 has a role in cell migration. a Over-expression of wild-type (1309S) and mutant (1309Y) TNS1. Wild-type and mutant TNS1 genes were constructed and stably transfected into the Caki-1 RCC cell line. Protein expression was assessed by western blot analysis. β-Actin was used as a positive control. b Mutant TNS1 has a role in cell migration. Cell migration was assessed in Caki-1 cells stably transfected with wild-type (1309S) or mutant (1309Y) TNS1 by a wound healing assay. The cells were cultured until confluent, then wounded with a pipette. Wound healing was observed at 24 h and 48 h later. Mock transfected cells were used as a negative control. c Mutant TNS1 does not have a role cell growth. Cell growth was assessed in Caki-1 cells stably transfected with wild-type (1309S) or mutant (1309Y) TNS1 by a cell count assay. The cells were seeded into 6-well plates, and cell numbers were counted after culture for 2 days. The data are shown as mean ± SEM (n = 3)
