a, b ELISA-based epitope mapping of mAbs with alanine scanned mutants. Utilizing ELISA, 32 mutants of NiV-RBP and 29 mutants of HeV-RBP were employed to evaluate the capacity of antibodies to bind, and the median effective concentration (EC₅₀) was calculated by fitting to a four-parameter curve using Prism software. Heatmap shows the fold-change in EC₅₀ binding titers of antibodies binding to henipavirus RBPs alanine mutants of compared with the wild-type proteins.
