Crypts are isolated from a surgically resected piece of neonatal intestine, and they are seeded in cell culture matrix hydrogel and grown into intestinal enteroids. Enteroids are dissociated and seeded in the top channel of the microfluidics device coated with extracellular matrix (ECM). Endothelial cells (HIMECs) are then added to the bottom channel.
