Graphical overview of experimental findings. Hearing loss, as a direct consequence of noise trauma is caused by loss of OHCs as well as synaptopathy. Our in vivo studies show compelling evidence that AC102 can prevent OHC loss, reverse synaptopathy, and subsequently restore hearing after acoustic exposure. Our in vitro experiments demonstrate neuroprotective and neuroregenerative properties of AC102 and revealed a reduction of ROS and increase of ATP as potential molecular mechanism that could explain the observed in vivo effects.
