aSplenocytes (2×105 /well) obtained from groups of immunized mice were cocultured in the presence of 1.0 μg of 3H1 or CEA in 96-well flat-bottomed microtiter plates in a final volume of 200 μl. Cell-free supernatants were harvested after 48 h (for quantitation of IL-2, IL-4, and IL-10) and 72 h (for quantitation of IFN-γ) of culture and cytokine levels were measured by standard ELISA kits.
