The SCSD-associated HAI-2 mutants cannot effectively suppress the prolonged lifespan of active prostasin caused by HAI-2 loss in Caco-2 cells. The four Caco-2 variants were treated with increasing concentrations of doxycycline (Dox.) to induce expression of wild-type HAI-2 (A) and the three SDSC-associated HAI-2 mutants (B, C, and D). Equal amounts of cellular proteins were analyzed by immunoblot for prostasin using the prostasin mAb YL11. The various prostasin species, including the complexes with the HAIs, active prostasin, and prostasin zymogen are indicated.
