Gq inhibition potentiates DAMGO inhibition of GABAergic eIPSCs in PAG slices. (A) Bar graph comparing effects of Gβγ and Gq inhibitors on inhibition of eIPSCs produced by a single dose of DAMGO (50 nM). All samples were preincubated with either vehicle control (DMSO) or the indicated inhibitors at 10 μM for 30 minutes, followed by addition of DAMGO. One-way ANOVA, F_{(3, 21)} = 14.4, P < 0.0001; Dunnett’s multiple comparisons, **P < 0.01, ****P < 0.0001. (B) Gallein (10 μM) and mGq-CT (10 μM) had no effect on DAMGO-dependent inhibition of eIPSCs in PAG slices isolated from PLCβ3^−/− mice.
