The HIV-2 Gag expression plasmid or selected mutant was transfected into 293T/17 cells, and the particles produced from the cells were concentrated and purified from cell culture supernatants prior to cryo-electron microscopy analysis. Representative images are shown for the (A) WT and the (B) G38M and (C) N127E CA domain mutants (scale bar, 150 nm). (D) Diameter quantification of the WT, G38M, and N127E immature particles (n > 50) generated by calculating the mean along two perpendicular axes per particle. The histogram shows the range, median, and the 25th to 75th percentiles of the particle diameters. Significance relative to WT was determined by an unpaired t test. ****, P < 0.0001.
