Secretion signal analysis. (A) Secretion efficiency of selected signal peptides. Mfr strains expressing several signal peptides with a luminescent reporter were grown to exponential phase. Luminescence of both supernatant (extracellular) and pellet (intracellular) was measured to obtain a secretion efficiency ratio. Data are shown as average +/- SD of two biological and technical replicates (N = 2). Statistical analysis was performed using One-way ANOVA test of multiple comparisons. The statistical analysis shown both compared to s3040 (*) and s515 (#) (p-value < 0.0002). (B) Luminescence signal of the supernatant in s3040 and s515. The signal was measured and normalised by quantifying the total protein in the pellet. Data are shown as average +/- SD of two biological and technical replicates (N = 2).
