(a) Vectors expressing gal4 DNA binding domain alone or fused to NRSF(1-73) were transfected into HEK293 cells along with vectors expressing TET-VP16 and the reporter plasmid shown (top). Values (bottom) are the averages of three independent transfections, each performed in triplicate with Tfx-50, normalized to cotransfected CMV-Renilla (see Materials and Methods), and expressed as a percentage of the value obtained with Gal4 alone. Standard errors were less than 10%. (b) Cells transfected as described were treated with 100 nM TSA for 24 h preharvest and analyzed as in panel a.
