Assembly of the TGEV full-length construct. (A) The various TGEV plasmid DNAs were digested with BglI, BstXI, or NotI, and the appropriate-sized products were isolated from agarose gels as described in the text. The TGEV A and B1 fragments, TGEV B2 and C3 fragments, or TGEV DE-1 and F fragments were ligated at 16°C overnight in separate reactions. Appropriate-sized products were isolated from agarose gels.
