PKCε is required for the binding of Smad3 to the promoter of glycolytic genes in prostate cancer cells. a The putative consensus sequence CAGACA of Smad3 binding element (SBE) and the number of the bases from transcriptional start codon (ATG, + 1) within promoter of the indicated target gene were analyzed by UCSC online software. b Smad3 binding to the promoter of glycolytic genes was determined in DU145 cells with or without PMA (100 nM) treatment using CHIP followed by RT-qPCR assay. c Smad3 binding to the promoter of glycolytic genes and Smad7 (positive control) were analyzed by CHIP assay in PC-3 cells infected with lentivirus GFP or PKCε in response to TGF-β1 (50 ng/mL) treatment. The ectopic expression of PKCε in PC-3 cells was confirmed by Western blotting (right panel).
