The statistical analysis of the number of gold grains of the 5mC immuno-gold labelling on condensed chromatin areas reveals a significant difference in the hepatocyte nuclei before and after the specific enzymatic digestion. The PK digestion significantly increases the immuno-gold labelling thus demonstrating not only that proteins do not contribute to the signal but also that the yield of the immuno-reaction could be increased removing chromatin associated proteins (p < 0.01). After the removal of both DNA and RNA the gold grains are significantly reduced on the condensed chromatin regions confirming that the signal depends on DNA (p < 0.01)
