Wnt5a stimulates the increase of the [Ca²⁺]_i mobilization in Ma-Mel-04 and Ma-Mel-12 melanoma cells, but not in HaCaT cell line. Untreated and ZAP- or pertussis toxin-treated cells were loaded with fura-2/AM and then 20 ng/ml of Wnt5a was added to start the reaction. Measurement of fluorescence by 340 and 380 nm excitation was performed each 60 s during 15 min, and the ratio 340/380 was calculated. For details, see “Materials and methods”
