Phosphatidylcholine reverses 5-LO inhibition by hyperforin. Partially purified human recombinant 5-LO (0.5 µg) expressed in E.coli was pre-incubated in 1 ml PG buffer plus 1 mM EDTA with hyperforin (a), ZM230487 (b), or BWA4C (c) at the indicated concentrations or with vehicle (DMSO) at 4°C with or without PC or PE (100 µg/ml, each) as given in the figure. ATP (1 mM) was added, and after 5–10 min, samples were pre-warmed at 37°C for 30 s and 5-LO product formation was started by addition of 2 mM CaCl2 and 20 µM AA. After another 10 min, 5-LO product formation was determined.
