Downregulation of reversion‐inducing‐cysteine‐rich protein with Kazal motifs (RECK) in human lung cancer cell lines and its restoration by 5′‐azacytidine. (A) Human lung fibroblast (1, MRC‐5) and lung cancer cell lines (2, H226; 3, H520; 4, H358; 5, A549) were harvested and subjected to isolation of genomic DNA, RNA and protein by TRIzol reagent. Reverse transcription–polymerase chain reaction (RT‐PCR) and western blot analysis were carried out to investigate RECK expression. (B) The methylation status of the RECK promoter was also addressed by methylation‐specific polymerase chain reaction. M, methylated; U, unmethylated. (C) H520 (3), H358 (4), A549 (5) and H226 (2) cells were treated with 5′‐azacytidine for 48 h and RECK expression was examined by RT‐PCR and western blot analysis.
