Generation of CD8⁺ CTL specific to OVA_257‐264 by vaccination with hsc70 fusion protein. C57BL/6 mice were immunized intraperitoneally with 10 µg OVA_265‐280‐hsc70‐OVA_257‐264, twice in a 1‐week interval. Two weeks after the second immunization, the spleen cells were stimulated in vitro with 10⁻⁶ M OVA_257‐264 for 5 days. (a, upper panel) The resulting cells were stained with tetramer consisting of H‐2K^b and OVA_257‐264 (MBL, Nagoya, Japan). (a, lower panel) CD8⁺ cells were sorted out by fluorescence‐activated cell sorting (FACS) Vantage. Cells in (a) were used for ⁵¹Cr‐release assay (b) and cytokine ELISPOT assay (c), as indicated.
