Effects of point mutations in the cytoplasmic tail of B5R on intracellular trafficking of chimeric proteins. HeLa cells were transfected with GFP chimeras containing the VSVG lumenal domain, the B5R transmembrane domain, and the B5R cytoplasmic domain with mutations in the putative retrieval sequences. Live, unpermeabilized cells were stained with VSVG MAb I1, followed by CY5-conjugated goat anti-mouse antibody to detect surface expression and Hoechst dye to show the locations of nuclei by confocal microscopy.
