Analysis of the effects of mutations in the Cys₃-His₁ motif of the M2-1 protein on interaction with the N protein. HEp-2 cells infected with vTF7-3 were transfected with pN alone (lane 1) or pN and a plasmid bearing a gene encoding the wild-type or a mutated M2-1 protein as indicated (lanes 2 to 6). Cells were exposed to [³⁵S]methionine and [³⁵S]cysteine for 2 h at 16 h posttransfection. Labeled proteins were immunoprecipitated from cytoplasmic extracts by using M2-1-specific MAb 5H5. Immunoprecipitated proteins were analyzed by SDS-PAGE in 11% polyacrylamide gels followed by fluorography. Positions of the M2-1 and N proteins are indicated. wt., wild type.
