Structure of ALK with two meprin A 5 proteins, receptor protein tyrosine phosphatase μ regions (MAM), glycine-rich domains (GR), a low-density lipoprotein motif (LDL), and an intracellular domain of tyrosine kinase [126]. Fusion with multiple possible partners leads to ligand-independent activation of downstream pathways, which are also activated under physiological conditions by other mechanisms. Activation of oncogenic pathways results in cancer initiation and progression by the mechanisms mentioned in the figure.
