Southern blot analysis of 8MA-V DNA. Total DNA harvested from Vero cells infected with KOS, 8MA, or 8MA-V was cleaved with PstI and XhoI. The resulting Southern blot was hybridized with a 2.8-kb PstI/XhoI fragment spanning the VP16 locus. 8MA-V displayed the 3.7- and 1.9-kb VP16 fragments indicative of the 8MA VP16 deletion-lacZ substitution mutation.
