Transient overexpression of UL26(M307L) and pre-VP21(M307L). Cells were transfected with plasmids expressing the indicated proteins and subsequently infected with the ΔICP35 virus, as described in Materials and Methods. In the left panel, cell lysates were examined for the expressed proteins by Western blotting with antisera against VP21/VP22a scaffold protein and the VP23 shell protein (for standardization of virus infection). The UL26/UL26.5 lane indicates expression of UL26 and UL26.5 under control of their native promoters from plasmid pRB4090 (29). For comparison, the right panel contains threefold dilutions of a wild-type-infected cell lysate. The Western blot was photographed using a Kodak DCS400 digital camera and Adobe PhotoShop 5.0.
