Expression pattern of REST and RCOR genes in human cerebral organoids derived from two healthy control iPSC lines (iPSC1 and iPSC2). (A) Cerebral organoids were matured for 9 months prior to harvesting. (B) Organoids were shown to be a mixed population of neurons (MAP2+) and astrocytes (GFAP+) via immunocytochemistry. Scale bar = 50 μm. (C) Organoid area (μm2) was measured over time with a steady increase in organoid size observed over maturation time. (D)REST and RCOR genes expression levels were analysed using RT-qPCR. Relative expression is calculated from the mean of three housekeeping genes and presented as mean ± SEM. 5–6 organoids were pooled together for each replicate, n = 3 biological replicates, n = 3 technical replicates. Data was analysed with a Two-way ANOVA with statistical significance determined using the Holm-Sidak multiple comparison method. *p < 0.05, **p < 0.01, ***p < 0.001.
