The ECS in ICP27 is required for nuclear retention of tsICP27. Vero cells were infected at an MOI of 10 with HSV-1, vBSLG4, or vBStsECS as indicated and incubated at 39.5°C. Infected cells were processed for immunofluorescence at 8 h postinfection following a 2-h treatment with cycloheximide. The intracellular distribution of ICP27 was examined with an antibody specific for ICP27 and an FITC-labeled secondary antibody.
