Pulse-chase analysis of ICP27 proteins synthesized in HSV-infected cells. Vero cells were either mock infected (M) or infected with HSV-1, vBSGFP27, or vBSGFPKH1 at an MOI of 10 and then incubated at 37°C. The cells were then pulse-labeled with Tran³⁵S-label for 15 min and either harvested (P) or chased by replacing the medium, incubated for 2 h, and then harvested (C). Whole-cell extracts were immunoprecipitated with an ICP27-specific polyclonal antibody, Clu38, and then subjected to SDS-PAGE analysis. Labeled proteins were visualized by autoradiography. Full-length ICP27 or GFP-ICP27 proteins are labeled as such. Molecular mass standards are indicated in kilodaltons.
