Entry of various HSV strains mediated by nectin-1α (FVB/N form). CHO-IEβ8 cells were transfected with nectin-1α-expressing pDS101 (hatched bars) or control vector pcDNA3 (open bars), replated in 96-well plates, and then inoculated with serial dilutions of the viruses indicated. After 6 h, the cells were permeabilized and incubated with ONPG substrate for quantitation of β-galactosidase activity. Entry of virus was signaled by induction of β-galactosidase expression from the lacZ gene in the CHO-IEβ8 cells. The values shown (means of triplicate determinations) represent the amount of reaction product detected spectrophotometrically (optical density at 410 nm [OD₄₁₀]) at a single input dose of 10⁶ PFU/well in the linear range of the dose-response curve for each virus. In this and other figures, each value shown is the mean of three or more determinations.
