Translation of monocistronic transcripts containing each possible combination of the HCV-H and HCV-N 5′NTRs and downstream coding sequences in rabbit reticulocyte lysates supplemented with increasing concentrations of KCl. Products of translation were separated by SDS-PAGE and quantified by PhosphorImager analysis. Relative band volumes were calculated based on the number of methionine residues in each of the two expressed polyproteins. pN-CΔE1 (···•···) and pH-CΔE1(H) (···▫···), or pN-CΔE1(H) (—•—) and pH-CΔE1 (—▫—), contain homologous or chimeric combinations of the 5′NTRs and polyprotein coding sequences of HCV-H and HCV-N, respectively.
