In vitro transcription with the hydrophobic-to-alanine mutant P proteins. (A) VVT7-infected A549 cells were transfected with no plasmids (−) or cotransfected with wt or mutant P plasmids together with L plasmid. Cell extracts were prepared and incubated with wt RNA-NP in the presence of [α-³²P]CTP. The transcripts were purified and analyzed by gel electrophoresis. The relative positions of the NP and P transcripts are indicated.
