Experimental absorbance spectra (red) of POPC vesicles diluted into isotonic or hypotonic buffers. The modeled absorbance (gray) has no fitting parameters and was determined using the expected sucrose concentration difference (Δc) between the interior and exterior of the vesicles. Taking the vesicle size distribution measured using DLS (black, dotted) into account makes little difference to the modeled absorbance.
