Slc30a10 Fpnfl/fl mice with and without Vil-ERT2 were injected intraperitoneally with a five-day course of tamoxifen starting at P14. Some mice were injected subcutaneously with Fe dextran on P19. At P28, mice were analyzed for: (a-e) liver (a), brain (b), bone (c), pancreas (d), and small intestine (e) Mn levels by ICP-OES; (f) blood Mn levels by GFAAS; (g) bile Mn levels by GFAAS; (h, i) Mn absorption by 52Mn gavage, with female and male data pooled in (h) and analyzed separately in (i). Data are represented as means +/− standard deviation, with at least four animals per group. Data were tested for normal distribution by Shapiro-Wilk test; if not normally distributed, data were log transformed. Within each sex, groups were compared using two-way ANOVA with Tukey’s multiple comparisons test. (* P<0.05, ** P<0.01, *** P<0.001, **** P<0.0001)
