Protein accumulation from the 5′ UTRs of 210 and 77 nt in vivo as a function of infection with BCV. (A) HRT-18 cells were infected with T7 RNA polymerase-expressing vaccinia virus at 1 h prior to transfection and then mock transfected or transfected with pCAT, p77(N)CAT, or p210(N)CAT plasmid DNA as schematically depicted; cells were then harvested 24 h later and assayed for CAT activity. Where indicated, cells were also infected with BCV at 5 h prior to transfection. Experiments were done in quadruplicate, and the standard deviations from the mean are indicated. (B) OST7-1 cells were infected with wild-type vaccinia virus at 1 h prior to transfection and transfected as indicated in panel A. Where indicated, cells were also infected with BCV at 5 h prior to transfection. Experiments were done in duplicate, and the ranges are shown.
